Introduction.

Stem cells play important role in etiology and development of the prostate diseases. CD133 antigen is one of the main markers for prostatic epithelial stem/progenitor cells. Doxazosin (Dox) induces apoptosis in both the prostatic epithelial and stromal cells.

Objectives.

The aim of study was to compare frequency of the apoptosis induced by doxazosin (Dox) in both stem/progenitor and adult cells of prostate epithelium.

Materials and methods.

Material for cultivate epithelial cells cultures, isolation and cultivate stem/progenitor cells cultures were from 10 BPH patients. After 14 days primary cultures were incubated for 12 hours with 20 (Dox 20), 50 (Dox 50) and 80 μM/l (Dox 80) of doxazosin. Cells viability was estimated with Trypan blue test as well as cells viability and frequency of apoptosis were estimated with Annexin V and propidium iodide test using flow cytometry.

Results.

90 cultures containing both the stem/progenitor cells (CD133(+)) and adult prostatic epithelial cells (CD133(-)) as well as 41 cultures containing sorted from prostate epithelium stem/progenitor cells (CD133(+)) were established. In CD133(+)/CD133(-) cultures after 12 hours of incubation with Doxazosin significant decrease of living cells percentage and significant increase of apoptotic cells percentage correlating with Doxazosin concetration compared with CD133(+)/CD133(-) cultures incubated without Doxazosin (control) were noted. In CD133(+) cultures there were no significant changes in living and apoptotic cells percentage after 12 hours incubation with Doxazosin comparing with control.

Conclusions.

Doxazosin induces apoptosis in adult prostatic epithelial cells. Proapoptotic influence on stem/progenitor prostatic epithelial cells is insignificant. That phenomenon may have influence on temporary effect of alpha1-antagonists in BPH treatment which is clinically observed.
